چکیده

Adenosine is a cellular metabolite with diverse derivatives that possesses wide range of physiological roles. We investigated the molecular mechanisms adenosine and cordycepin for their promoting effects in wound-healing process. The mitochondrial energy metabolism cell proliferation markers, cAMP responsive element binding protein 1 (CREB1) Ki67, were enhanced by cultured dermal fibroblasts. stimulated receptor signaling via elevated cAMP. phosphorylation mitogen-activated kinase (MEK) 1/2, mammalian target rapamycin (mTOR) glycogen synthase 3 beta (Gsk3b) Wnt genes such as bone morphogenetic (BMP) 2/4 lymphoid enhancer factor (Lef) activated. gene expression was abrogated A2A A2B inhibitors, ZM241385 PSH603, A (PKA) inhibitor H89, indicating involvement A2A, PKA. As result Wnt/?-catenin pathway activation, secretion growth factors insulin-like (IGF)-1 transforming (TGF?) increased, previously reported to facilitate wound healing In addition, vitro fibroblast migration also demonstrating possible roles facilitating conclusion, our data strongly demonstrate stimulate through activation receptor, possibly tissue remodeling process suggest therapeutic potential treating skin wounds.

Adenosine augments IL-10 production by microglial cells through an A2B adenosine receptor-mediated process.

Microglia are activated by pathogen-associated molecular patterns and produce proinflammatory cytokines, such as TNF-α, IL-6, and IL-12, and the anti-inflammatory cytokine IL-10. Adenosine is an endogenous purine nucleoside and a ligand of four G protein-coupled adenosine receptors (ARs), which are the A(1)AR, A(2A)AR, A(2B)AR, and A(3)AR. ARs have been shown to suppress TNF-α production by mic...

Methylthioadenosine Reprograms Macrophage Activation through Adenosine Receptor Stimulation

Regulation of inflammation is necessary to balance sufficient pathogen clearance with excessive tissue damage. Central to regulating inflammation is the switch from a pro-inflammatory pathway to an anti-inflammatory pathway. Macrophages are well-positioned to initiate this switch, and as such are the target of multiple therapeutics. One such potential therapeutic is methylthioadenosine (MTA), w...

The Canonical Wnt Signaling (Wnt/β-Catenin Pathway): A Potential Target for Cancer Prevention and Therapy

Precise regulation of signal transduction pathways is crucial for normal animal development and for maintaining cellular and tissue homeostasis in adults. The Wnt/Frizzled-mediated signaling includes canonical and non-canonical signal transduction pathways. Upregulation or downregulation of the canonical Wnt-signaling (or the Wnt/β-Catenin signal transduction) leads to a variety of human diseas...
